Changing text using includes.php

In the inc/languagefile_en.php file, text is this way [code]$GLOBALS['xx34code']='3 or 4 digit code from back of card';[/code] But when I make changes in the includes.php file, I always do it this way [code]$xx34code="3 or 4 digit security code";[/code] Simply question - does the format matter in the includes.php file?
